E1A基因对人宫颈癌细胞化疗药物敏感性的影响 被引量:3

摘要 目的探讨E1A基因对人宫颈细胞化疗药物的增敏作用及其相关机制。方法E1A基因经PEI-Fe3O4纳米粒介导转染人宫颈癌细胞系,用不同浓度的顺铂、5-氟尿嘧啶和紫杉醇处理细胞,通过MTT法测绘细胞存活曲线,观察E1A基因对顺铂、5-氟尿嘧啶和紫杉醇等化疗药物敏感性的影响;用同一浓度的顺铂、5-氟尿嘧啶和紫杉醇处理细胞,观察E1A基因在不同时间对癌细胞存活率的影响。结果转染E1A基因的人宫颈癌细胞(Hela-E1A)对顺铂和紫杉醇的敏感性显著增加,与Hela和Hela-vect细胞系比较,Hela-E1A细胞对顺铂和紫杉醇的敏感性分别提高了10倍和15倍;对5-氟尿嘧啶的增敏作用不明显。结论E1A基因能显著提高人宫颈癌细胞对顺铂和紫杉醇等化疗药物的敏感性。其作用机制可能是E1A基因抑制了该细胞的HER-2/neu基因的表达,使该细胞周期再分布,出现S期抑制和G2/M期阻滞。 [Objective] To investigate sensitization of chemotherapeutics drug to human cervical carcinoma cell and correlated mechanism. [Methods] The cell line transfected with E1A gene was treated by cisplatin, 5-fluorouracil and paclitaxel respectively, with different saturation to observe the effect of E1A gene on the sensitivity of the chemotherapeutics drug such as cisplatin, 5-fluorouracil and paclitaxel by MTT and those with the same saturation to observe the effect of E1A gene on the cell survival rate in different time. [Results] Human cervical carcinoma cell transfected by E1A gene was remarkably more sensitive to cisplatin and paclitaxel, but not sensitive to 5- fluorouracil and the sensitivity was increased by 10 times and 15 times compared with that of Hela cells and Helavect cells. [Conclusions] E1A gene can effectively enhance the sensitivity of human cervical carcinoma cell to the chemotherapeutics drug such as cisplatin, and paclitaxel excluding 5-fluorouracil. The mechanism of action is probably that E1A gene suppress the expression of HER-2/neu gene of the cell which changed the redistribution of the cell cycle and leaded to the suppression of S stage and the blockage of G2/M stage.
