Calpain重组分子免疫小鼠IFN-γ和IL-4表达的变化 被引量:1

摘要 目的探讨日本血吸虫疫苗侯选分子—钙离子激活的中性蛋白激酶(Calpain)的保护性免疫和免疫保护机制。方法用重组纯化的Calpain抗原免疫BALB/c小鼠,RT-PCR分析小鼠—氧化氮激酶(iNos.enos.nNos)mRNA的表达;ELISA分析免疫小鼠体外培养脾细胞产生的细胞因子IFN-γ和IL-4的动态变化。结果重组Calpain抗原免疫小鼠1周后,小鼠iNosmRNA的表达显著增强;培养24h免疫鼠脾细胞上清中IFN-γ浓度显著高于对照组,而IL-4的产生在免役组与对照组之间无显著性差异。结论Calpain抗原能诱导IFN-γ的产生,提示Calpain诱导Th1的免疫应答反应来抗日本血吸虫的感染。 Objective To explore the protective immunity and mechanisms of Calpain, a calcium - activated meutral proteinase as a vaccine candidate molecule. Methods Anti - Calpain sera was prepared by immunizing ALB/c mice with recombinant purified Calpain and mRNA expression of iNos, nNos and eNoswere detected by RT- PCR, and the concentration of of IFN- γ and IL- 4 in spleen cell of culture of immunized mice were analyzed by using ELISA. Results High level of IgG antibody specific to the antigen was observed in mice immunized with recombinant calpain. The concentration of IFN - γin the supernatant of mouse spleen cell 24 hours after culture was significantly higher than that of control, while no significant differences were noticed in the production of IL - 4 in the immunized mice as compared with that of the control. Conclusion Calpain can induces the production of IFN - γ, suggesting that the immune response of Thl induced against infection with Schistosoma japonicum.
