摘要
目的:观察酪丝亮肽对人肝癌BEL-7402的抑制作用,并分析其可能机制。方法:制备裸鼠移植瘤模型,随机分为生理盐水、氨基酸混合物、环磷酰胺、酪丝亮肽高低剂量等5组,每组12只,观察酪丝亮肽对人肝癌BEL-7402肿瘤组织坏死和凋亡的影响;电镜观察其对肿瘤细胞超微结构改变;流式细胞技术分析其对细胞周期及细胞凋亡率的影响。结果:酪丝亮肽能够诱导肿瘤细胞坏死和凋亡、细胞周期阻抑、显著抑制肿瘤的生长,给药剂量为80μg.kg-1.d-1和160μg.kg-1.d-1时,肿瘤抑制率分别为42%与58%。结论:酪丝亮肽可以显著抑制人肝癌BEL-7402的生长,作用机制与促进肿瘤细胞的凋亡和坏死有关。
AIM : To investigate the antitumor effect of tyroserleutide on the growth of the human BEL-7402 hepatocarcinoma inoculated into nude mice and explore preliminary mechanism. METHODS:The nude mice bearing xenografts of the human BEL-7402 hepatic carcinoma were given daily ip injections of tyroserleutide or saline or mixture of amino as controls after tumor implantation, altogether about five groups with twelve mice for each. Apoptosis and necrosis were determined with fluorescence-activated cell sorter (FACS) and electron microscopy for detecting cellular microstructural charges. could significantly induce growth in nude mice and RESULTS : Tyroserleutide the inhibition tumor of changed the ultrastructure and cell cycle of tumor cell. At dose of 80 μg·kg^-1·d^-1and 160 μg·kg^-1·d^-1 it could significantly inhibit tumor growth in nude mice by inhibition rates of 42 % and 58 % respectively. CONCLUSION : Tyroserleutide can inhibit significantly tumor growth in nude mice. The growth inhibition of the xenografts may concern with the apoptosis of tumor cells induced by tyroserleutide.
