摘要
目的研究普洛迪注射液对大鼠局灶脑缺血再灌注性损伤的保护作用。方法采用Zea longas法制备大鼠脑缺血再灌注(MCAO/R)模型,分为假手术组、缺血再灌注组、普洛迪注射液药物干预组。各时相点取材,HE染色观察镜下改变,测定脑含水量;免疫组化观察脑组织血管内皮生长因子(VEGF)的表达变化,并使用多媒体彩色病理图像分析系统进行定量分析;利用电镜技术观察脑组织超微结构改变。结果普洛迪注射液药物干预组与其余2组对比,脑含水量及VEGF的表达均有显著性差异。结论普洛迪注射液可能减少缺血再灌注后脑含水量并增强VEGF表达,减轻缺血再灌注性脑损伤,从而起到神经保护作用。
Objective It is to study the protective action of Puluodi injection on focal cerebral ischemia reperfusion in rats. Methods Cerebral ischemia reperfusion rat models were prepared with Zea longas method and randomly divided into pseudooperation group, ischemia reperfusion group and Puluodi injection intervention group. Pathological changes were observed with HE staining and transmission electron microscope. The content of brain water was assayed. The expression of VEGF was observed with immunohistochemical method and quantitative analysis was carried out with multimedia color pathological image analysis system. Results There were obvious differences on the content of brain water and the expression of VEGF between Puluodi injection intervention group and the other two groups. Conclusion Puluodi injection may decrease content of brain water, enhance the expression of VEGF after ischemia reperfusion and lighten cerebral ischemia reperfusion injury to actualize neuroprotective effects.
