摘要
目的建立反相高效液相色谱法测定齐墩果酸原料及片剂中齐墩果酸含量的方法,同时利用二极管阵列检测器建立了齐墩果酸峰纯度的鉴定方法. 方法色谱柱为Nova-Pak C18,流动相为乙睛-水(65∶ 35),流速0.8 ml·min-1,检测波长210 nm,柱温25℃.结果齐墩果酸在3.48~17.40 μg范围内线性关系良好(r=0.999 9),检出限为0.3 mg·L-1 (3σ),平均回收率为99.5%. 结论该方法简便,准确,线性范围宽,重现性好,适于对含有该成分的药物进行质量评价.
Aim Adetermination method of oleanolic acid in material and oleanolie acid troches by reversed-phase high performance liquid chromtograph was established and at the time the purity of oleanolic acid was derected by means of PhotodiodeArray Detector. Methods The chromatographic column, Nova-Pak C18 (3.9 150 mm, 4 λm), ethanenitrile-water (65: 35,V/V) mobile phase with 0.8 ml·min^-1 flow rate, the detected wavelength (210 nm) , and the column temperature (25 ℃ ) were adopted. Result The calibration curve of oleanolic acid was linear under the content of 3.48 -17.40 μg,the correlation coefficient was over 0. 9999, the detected limit was 0.3 ml·L^-1 ( 3 or), The average recovery of oleanolic acid was 99.5 %. Conclusion The method is simple, accurate and has wider linear range. The method can be adapted to evaluate the quality of medicine that contain oleanolic acid.
