不同年龄大鼠骨髓基质干细胞的增殖特性

摘要 为了探讨年龄差异是否对大鼠骨髓基质干细胞(BMSCs)的增殖有影响。本研究采用免疫组织化学和苏木素染色检测体外培养的不同年龄大鼠BMSCs增殖能力的变化。结果显示:在培养的第5d,新生组增殖率为(78.7±5.3)%,成年组为(72.0±3.3)%,老年组为(65.4±3.9)%;在第7d增殖率分别是(78.3±3.2)%、(67.3±3.9)%和(52.6±6.0)%;在第9d,分别是(66.9±3.3)%、(46.4±3.9)%和(39.1±2.2)%。统计分析表明,在培养的第5、7、9d三个年龄组BMSCs的增殖率均有显著性差异(P<0.05)。结果提示:不同年龄的骨髓基质干细胞在体外均能快速增殖,但随着年龄的增大,其增殖能力明显下降。 To investigate the effect of age ou the proliferation capability of bone marrow stromal cells (BMSCs). Immunohistochemistry and hematoxylin staining methods were used. At day 5 of cell culture, the proliferation rates of BMSCs from neonatal, adult and aged rats were(78.7±5.3)%, (72.0±3.3)ck and (65.4±3.9)%, respectively. At day 7, they were(78.3±3.2)%, (67.3±3.9)% and (52.6 ±6.0)%, while at the 9th day, they were down to (66.9 ±3.3)%, (46.4 ±3.9)% and (39.1 ±2.2)%. Statistical analysis showed that there was a significant difference in the culture of day 5,7 or 9 among three groups ( P 〈0.05). The results suggest that BMSCs are able to proliferate quickly but the capability decreased with age.
