摘要
目的:就关于合成各种前列腺素的关键酶环氧合酶2在中枢神经系统生理和病理过程中的作用的研究进展进行概括总结。资料来源:应用计算机检索Medline1995-02/2005-02的与环氧合酶2的神经生物学作用相关的文章,检索词“COX-2,brain,CNS”,并限定文章语言种类为English。同时计算机检索CNBI1995-02/2005-02期间相关的文章,限定文章语言种类为中文,检索词“环氧合酶2,脑,中枢神经系统”。资料选择:就检索到的280余篇文献进行筛选,选择以环氧合酶2在神经系统生理和病理过程中的作用为主要研究内容的文献35篇,无论研究对象为动物还是患者全部纳入,其中研究内容相似的,以近3年且发表在较权威杂志者优先。资料提炼:将筛选到的文献按照不同生理过程或者病理过程分类,其中20篇与不同的病理过程相关,7篇与环氧合酶2的生理功能相关,其他还有一些与环氧合酶2在神经系统以外的作用相关。资料综合:35篇文献共包括近千例实验动物(或患者),说明了环氧合酶2参与兴奋性神经元的突触传递、中枢神经系统的炎性过程、痛觉过敏和各种脑损伤过程,也参与神经变性病如阿尔茨海默病和帕金森病的病理过程等。结论:环氧合酶2在神经系统的生理和病理过程中具有重要的作用。对其神经生物学作用的深入研究将为神经系统许多疾病的临床治疗提供新的思路。
OBJECTIVE: To sum up the research development of effects of cyclooxygenase-2 (COX-2) that is the key of synthesizing various kinds of prostaglandin on physiological and pathological procedures of central nervous systenl. DATA SOURCES:We searched Medline database for the related articles on neurobiology functions of COX-2 published between February 1995 and February 2005, with the key words “COX-2, brain, CNS” by computer and the language was limited to English. At the same time, the related articles were researched in CNB1 published February 1995 and February 2005, with the language limited to Chinese, and with the key words “COX-2, brain, central nervous system”. STUDY SELECTION: The over selected 280 articles were screened, and 35 articles, which took functions of COX-2 in physiological and pathological procedures of central nervous system as the main study contents, were selected. In the studies the experimental subjects included animals and patients. If the study contents were similar, the more authoritative ones that were published in recent 3 years were selected. DATA EXTRACTION: The articles selected were classified according to different physiological and pathological procedures. Twenty out of the articles were relevant to different pathological procedures. Seven articles were about the physiological functions of COX-2, The others were about COX-2 effects out of central nervous system. DATA SYNTHESIS: The 35 articles which included about 1 000 experimental animals (or patients) showed that COX-2 participated in the synaptic transmission of the excitatory neurons, inflammation procedure in central nervous system, hyperalgesia, various kinds of brain injuries procedures and neural degeneration diseases such as the pathological procedures of the Alzheimer disease and Parkinson disease etc. CONCLUSION: The COX-2 plays important roles in the physiological and pathological procedures of central nervous system, The further study about neurobiology effects of COX-2 would provide new clues for clinical treatment of many diseases in nervous system.
