波生坦对单侧输尿管梗阻大鼠的肾脏保护作用

摘要 目的探讨波生坦对单侧输尿管梗阻(UUO)大鼠肾脏病理改变的影响。方法雄性Wistar大鼠.随机分为假手术组(S)、手术组(UUO)和波生坦组(Bos),各12只。制备UUO大鼠模型。Bos组给予波生坦100mg/(kg·d)灌胃.术后4个时间点观察血常规、肾脏大体和组织病理。免疫组织化学测定肾组织转化生长因子-β1(TGF-β1)表达.放射免疫法测定血浆、梗阻侧肾脏残尿内皮素-1(ET—1)浓度。结果梗阻侧肾脏大体观为体积增大,肾盂扩张,肾皮质变薄。TGF-β1阳性细胞数表达增多,组织病理改变,显示间质纤维化。结论波生坦能减轻UUO大鼠肾间质纤维化程度,其可能通过ET和TGF-β1机制起作用。 Objective To research the effects of bosentan in on unilateral ureteral obstruction (UUO) rats. Methods Wistar rats were randomly divided into three groups. UUO group was subjected to complete ligation of left ureter, shanm operation group with sham operation was used as control, Bosenten group was subjected to complete ligation of left ureter and to stonmch with bosentan suspension. Three groups were killed at 5,10,15,20 d. We examined renal pathological changes and the expressions of transforming growth factor-β1 (TGF-β1 ) in kidney of all rats using immunohistochemistry analisis method. The plasma and ureteral obstructive kidney urinary concentration of endothelin - 1 ( ET - 1 ) was determined by radioimmunoassay. Results In Boss and UUO groups, the urinary concentration of ET - 1 increased substantially and the expression of TGF -β1 were much higher than those in the S group. At the same time, tubulointerstitial fibrosis was present in the kidney of rats in Bos and UUO group. But the expression of TGF -β1 and tubulointerstitial fibrosis in Bos group were lower than those of UUO group. Conclusion Bosentan can effectively amelirate tubulointerstitial fibrosis in rats with UUO by ET and TGF -β1 machanism.
