摘要
应用聚丙烯酰胺凝胶等电聚焦电泳技术,对雄激素受体亚群进行了研究。结果发现,前列腺增生组织中有三种雄激素受体亚型,等电点(PI)分别在6.5、6.0和5.3。其中95%(19/20)的标本含有P16.5、5.3亚型,45%(9/20)的标本含有PI6.5、6.0和6.3亚型,50%(10/20)的标本含有PI6.0亚型,5%(1/20)的标本只有P15.3亚型。3例前列腺癌标本中,2例有P16.5、6.0、5.3三种亚型,1例有6.5、6.5两种亚型。氘标的双氢睾酮(DHT)与三种受体亚型的结合,可以被非氚标的DHT,睾酮(T)竞争抑制,而孕酮、雌二醇和己烯雌酚对此却无竞争抑制作用。
With isoelectric focusing technique,the heterogenous nature of androgen receptors(AR)in benign hyperplastic and inalignant prostatic tissues was studied,AR isoforms could be detected at PI values of 6.5,6.0 and 5.3.In 19 of the 20 samples,the isoform was detected at PI 6.5 and 5.3;9 of the 20 expressed all the isoforms;10 of the 20 expressed the PI 6.0 isoform and 1 expressed the PI 5.3 isoform. 2/3 of the malignant tissues contained all the 3 isoforms and 1/3 contained the PI 6.5,6.0 isoforms. Binding of(3H)dihydrotestosterone to the 3 isoforms could be inhibited by 100 fold excess of dihydrotestosterone and testosterone but not by progesterone,oestradiol of diethylstibaestrol.Genesis of these isoforms has not yet been clear but they may play an important role in tissue sensitivity to the androgens.
