摘要
目的研究缺氧缺血新生猪凝血状态,以及部分凝血因子促凝活性的动态变化,探讨缺氧缺血性脑损伤(HIBD)时凝血功能变化。方法制作新生猪HIBD模型,并设对照组。于恢复正常供氧后0h, 24h,48h及72h时,测定血浆活化部分凝血活酶时间(APTT),凝血酶原时间(PT),凝血酶时间(TT)及凝血因子Ⅱ,Ⅴ,Ⅸ,Ⅺ的促凝活性。结果在HIBD后48h,APTT,PT,TT均较对照组明显缩短,差异有显著意义(P<0.05);凝血因子Ⅱ,Ⅴ促凝活性较对照组显著升高(P<0.05)。凝血因子Ⅸ,Ⅺ促凝活性,在 HIBD后48h均数最高,有上升趋势,结论在HIBD中存在凝血功能改变,在HIBD后48h变化最显著, 血液处于高凝状态,可进一步加重脑缺氧缺血。这是新生儿缺氧缺血性脑病(HIE)发病机制中“脑血流改变”的致病因素之一。
Objective To investigate the changes of blood coagulation state and some prothrombin's function in newborn piglets at different time after hypoxic-ischemic brain damage(HIBD). Methods The study was performed on newborn swines(n=40), in which the left carotid was ligated and the animals were treated under hypoxia(8% oxygen)for two hours. APTT,PT,TT and the activities of prothrombin II, V, IX and XI were measured at once and at 24,48,72 hours respectively after hypoxia and compared with the control group. Results APTT, PT and TT shortened and the activities of prothrombin II,V elevated obviously at HIBD 48h (P〈0.05). The activities of prothrombin IX and XI rose to the highest level at HIBD 48h. Conclusion In neonatal swings with HIBD, the blood coagulation state changed. At HIBD 48h, the piglets were at high coagulability,which may be related to the brain infarction in HIE.
