摘要
由根霉PW 358经固态发酵生产脂肪酶,以此酶为催化剂,在非水介质中合成了丁酸异戊酯。该文研究了反应温度、溶剂、底物浓度、底物摩尔比、吸水剂用量等因素对酯化反应的影响,确定了丁酸异戊酯的最佳合成条件为:反应温度为50℃,正庚烷为反应介质,丁酸与异戊醇的摩尔比为1∶1.2,两底物浓度均为0.25 mol/L。在反应体系中,每毫升溶液需加入0.25 g的5分子筛,以吸收酯化反应生成的水,促使平衡向酯化方向移动。在优化的条件下,反应5 h后,合成丁酸异戊酯的转化率达97%。
The Rhizopus sp. PW358 lipase catalyzed synthesis of isoamyl valerate in organic solvents was studied, and the optimum condition for the esterification was acquired. The esterification reaction was carried out in n-heptane solvent at 50 ℃, with 0. 25 mol/L of substrate concentration and 1: 1.2 of acid-alcohol molar ratio. The 5 A° molecular sieves were added to the reaction system as water adsorbent with 0. 25 mg/mL in the system. Under this condition, esterification conversion rate reached 97% after incubation for 5 hours.
