淋球菌RAPD分型的引物筛选与应用

摘要 目的为了研究不同淋球菌菌株的基因多态性,了解不同菌株之间的亲缘关系,选择适当的随机引物。方法以淋球菌基因组DNA为模板,对淋球菌进行RAPD扩增,选择多态性好、重复性好、稳定性强的随机引物,并用S38号引物对不同淋球菌基因组DNA进行指纹图谱区分。结果从Sagon公司50条RAPD随机引物中筛选出8条扩增效果良好的引物,经RAPD扩增出的淋球菌指纹图谱间有明显不同的DNA多态性,可对不同菌株进行聚类分析。结论利用RAPD技术对淋球菌进行随机引物的筛选,可为今后淋球菌的分子流行病学研究奠定基础。 Purpose In order to investigate the genetic diversity within different NG strains and screen the random primers. Methods In order to select applicable random amplified polymorphic DNAs(RAPD) primers for Neisseria gonorrhoeae genotyping,different RAPD primers sequence were used for amplification and their differentiating capability for Neisseria gonorrhoeae strains were compared;fingerprint map of Neisseria gonorrhoeae strains were produced by the primer S38. Results Eight suitable primers were screened from total 50 random primers; the fingerprint map of different Neisseria gonorrhoeae strains showed obviously polymorphic,and could be used to classify different Neisseria gonorrhoeae strains. RAPD could get the suitable primers and set the base for research in the future.
