刺五加胶囊对抑郁模型大鼠结肠组织iNOS、NO和COX-2的影响 被引量:1

摘要 ①目的探讨刺五加胶囊对抑郁模型大鼠结肠组织的作用及其机制。②方法建立大鼠抑郁模型,予以刺五加胶囊(剂量300、600、900mg/kg)与5氨基水杨酸(5ASA,100mg/kg)灌肠给药2周后,采用生化方法检测大鼠结肠组织中诱导型一氧化氮合酶(iNOS)和一氧化氮(NO)的含量;用免疫组化方法检测结肠组织环氧合酶2(COX2)的表达。③结果抑郁模型大鼠结肠组织中iNOS和NO的含量均明显升高(F=21.266、11.741,q=5.543~9.841,P<0.01),COX2在抑郁大鼠结肠中表达增加(F=51.079,q=7.843,P<0.01);各剂量刺五加胶囊灌肠后iNOS和NO的含量降低(q=7.726~8.902,P<0.01),600、900mg/kg刺五加胶囊灌肠后COX2的表达明显降低(q=9.716~11.402,P<0.05)。④结论刺五加胶囊可能通过下调抑郁模型大鼠结肠组织iNOS和NO含量及COX2表达发挥对结肠的保护作用。 Objective To investigate the protective effects of Ciwujia capsule on the colon of depression model rats and its mechanisms. Methods The depression model of rat was produced. Ciwujia capsule and 5-ASA were administered by enema for two weeks. The inducible nitric oxide synthase (iNOS), nitric oxide (NO) and the expression of cyclooxygenase-2 (COX-2) in the colon were detected by corresponding kits and immunohistochemical technology. Results Depression increased significantly the levels of iNOS, NO and the expression of COX-2; Ciwujia capsule (300,600,900 mg/kg)could down-regulate the levels of iNOS and NO. At dose of 600,900 mg/kg ,the capsules could decrease the expression of COX-2. Conclusion Ciwujia capsule can protect the colon of depressed rats by decreasing the levels of iNOS, NO and the expression of COX-2 in colon.
