摘要
目的测定复方补骨脂酊中补骨脂素和异补骨脂素的含量。方法采用YWGC18色谱柱(4.6mm×200mm),流动相为54%甲醇(含1%冰醋酸溶液),流速为0.7ml/min,检测波长为295nm。结果补骨脂素和异补骨脂素保留时间分别为9.165min和10.082min,标准曲线的线性范围均为20~200mg/l(补骨脂素r=0.9999;异补骨脂素r=0.9993)。补骨脂素和异补骨脂素中的高、中、低、三种浓度溶液的日间和日内变异均小于10.0%,加样回收>90%。测定复方补骨脂酊中补骨脂素和异补骨脂素的含量分别为2.058、2.270、2.201mg/ml和1.665、1.617、1.609mg/ml。结论本法简便、快速重现性好,可用于复方补骨脂酊制剂质量控制的含量测定方法。
Objective HPLC method for determination of the contents of psoralen and isopsoralen angelicin m compound Buguzhi tincture was employed. Methods The HPLC analysis was made on a column of YWG C18(4. 6 mm× 200 mm)with a mobile phase of 54 % methanol (1% acetic acid glacid solution contained). The detection wavelength was at 295 nm and the flow rate was 0. 7 ml/min. Results The retention time of psoralen and isopsoralen angelicin were 9. 165 min and 10. 082 min respec- tively. The linearity range of psoralen (r= 0. 9999)and isopsoralen angelicin (r= 0. 9993) were between 20 to 200 mg/l. The withday and between-day RSD for psoralen and isopsoralen angelicin at different concentration was less than 10. 0% with average recovery rate more than 90%. The content of psoralen and isopsoralen angelicin in compound Bugnzhi tincture was 2. 058,2. 270, 2. 201 mg/ml and 1. 655,1. 617,1. 609 mg/ml respectively. Conclusion The method is simple and reproducible and may be taken as the quality control for compound Buguzhi tincture.
