摘要
本试验以盆栽３年生新红星／海棠为试材，研究了苹果新根细胞分裂素的提取、纯化和高效形相色谱（ＨＰＬＣ）分析方法测定。苹果新根用８０％冷甲醇抽提，在ｐＨ３．０下用石油醚、乙酸乙酯和在ＰＨＳ．０～８．５下用正丁醇萃取，提取滚浓缩后用ＰＨ３．０的磷酸缓冲液做溶剂，加ＰＶＰ振荡吸附，最后用ＳｅＰ—ＰａｋＣ１８简柱进行纯化，以甲醇、水溶剂系统（ＰＨ３．５）在ＳｐｈｅｒｉｓｏｒｂＣ１８柱上进行分离，用紫外检测器在２６５ｍｍ波长下检测、鉴定．苹果新根中的ＣＴＫ种类主要有Ｚ、ＺＲ、ｄｉＨＺ。
Abstraction and purification of endogpnous cytokinins in new apple roots and their efficiently analytical methods of liquid chromatography were studied. Three-year -old pot-cultured apple(starkrimson/m. prunifolia) was used as material for the experiment. The results were as follows: 1.cytokinins were extracted from roots with 80% cold methanol,initially purified with petroleaum and n-BuoH. Insoluble polyvinylpyrvolidone(PVP)and C18cartridges were used for further purification,and finally separated and quantified with methanol-water on Spheerisorb C18 reversed phase HPLC columns.2.Cytokinins in apple roots had two types:both zeatin and zeatin riboside.
