氟尿嘧啶区域动脉灌注对重症急性胰腺炎大鼠细胞凋亡的影响 被引量:1

The Effect of Regional Intra-arterial Infusion with Fluorouracil on Apoptosis in Rats with Severe Acute Pancreatitis

摘要 【目的】探讨氟尿嘧啶区域动脉灌注对重症急性胰腺炎(SAP)大鼠细胞凋亡的影响。【方法】SD大鼠64只,随机分为假手术组(A组),SAP组(B组),氟尿嘧啶外周静脉注射组(C组),氟尿嘧啶区域动脉灌注组(D组)。测定各组大鼠的血清淀粉酶(Amy),观察胰腺病理变化,并检测胰腺腺泡细胞的凋亡率。【结果】①制成SAP模型后,各组Amy水平显著上升,氟尿嘧啶治疗后可使Amy下降,其中氟尿嘧啶区域动脉灌注组下降更为明显。②氟尿嘧啶治疗可使胰腺腺泡细胞凋亡率明显上升,胰腺病理损害程度减轻,氟尿嘧啶区域动脉灌注者作用更强。【结论】①SAP时,应用氟尿嘧啶可诱导胰腺细胞凋亡,减轻胰腺病理损害,是治疗SAP的方法之一。②区域动脉灌注是治疗SAP的增效途径。 [Objective]To study the effects of regional intra-arterial infusion with fluorouracil (Fu) on apoptosis in rats with severe acute pancreatitis (SAP). [Methods]SD rats were divided into four groups randomly: Sham-operative group (group A), SAP group (group B), group of peripheral vessel infusion of Fu (group C), group of regional intra-arterial perfusion of Fu (groupD). The contents of serum amylase and the pancreatic histologic changes were observed and apoptosis of pancreatic acinar cells was detected by triphosphate-biotin nick-end Labelling (TUNEL). [Results]①The serum amylase level of each group after establishment of model was much higher than before, treatment with Fu decreased the serum amylase level, regional intra-arterial perfusion of Fu had the most remarkable effectiveness. ②Treated with Fu, the apoptotic ratio of acinar cells improved, the pathological damage severity of pancreas was significantly attenuated, especially in group D.[Conclusion]①During the course of SAP, treatment with Fu could improve the apoptotic ratio of acinar cells, has beneficial effect on pancreas damage.②Regional intra-arterial perfusion is one of the means of improving therapeutic effect of drugs against SAP.
