八肽胆囊收缩素对体外胰岛素β细胞增殖及胰岛素分泌的影响 被引量:1

摘要 目的研究八肽胆囊收缩素(cholecystokinin,CCK-8)对小鼠胰岛β细胞(NIT-1细胞)增殖及胰岛素分泌的影响.方法体外培养NIT-1细胞,分别加入10-8~10-10mmol/L浓度CCK-8处理24和48 h,MTT法检测NIT-1细胞增殖情况;放免法检测用药后NIT-1细胞胰岛素分泌量.结果与对照组比较,经不同CCK-8处理24及48 h后均可显著提高NIT-1细胞的增殖及胰岛素分泌量,且具有剂量及时间依赖性.10-8 mmol/L CCK-8组与对照组相比,可提高增殖率分别为24 h 48%,48 h 77%;与对照组比较,10-8 mmol/L CCK-8组可增加胰岛素释放量分别为24 h 181%,48 h 151%.结论 CCK-8具有显著增强NIT-1胰岛β细胞增殖及胰岛素分泌的作用. Objective To study effect of cholecystokinin-octopeptide (CCK-8) on isletβcell line NIT-1 proliferation and insulin secretion. Methods In vitro cultured NIT-1 cells were exposed to CCK-8 in a dose of 10^-10- 10^-8 mmol/L for 24 and 72 h,NIT-1 cell proliferation was detected by MTT. NIT-1 cell insulin secretion after exposed to CCK-8 were determined by radioimmunity. Results NIT-1 cell proliferation and insulin secretion increased 24 and 48 h after exposed to CCK-8 with dosage-time dependency comparing to those in control. CCK-8 in dose of 10^-8 mmoL/L increased the cell proliferation rate 48% at 24th hour and 77% at 48 h comparing to those in control and enhanced insulin secretion 181% at 24 h and 151% at 48 h. Conclusions CCK-8 could play promotion role in NIT-1 insulinβcell proliferation and insulin secretion.
