摘要 目的研究柴油车尾气颗粒物有机提取物(DEPE)对中国仓鼠肺成纤维细胞(V79)的DNA损伤及作用途径。方法通过单细胞凝胶电泳试验检测DEPE致细胞DNA单链、双链断裂及DNA蛋白质交联水平,并用抗氧化剂N乙酰半胱氨酸(NAC)作干预试验,探讨其损伤机制。结果DEPE能致细胞DNA单链及双链断裂,在500~1000μgmL能引起一定程度的DNA蛋白质交联,NAC可降低DEPE致DNA损伤。结论在一定剂量下,DEPE可致细胞DNA单链、双链断裂及DNA蛋白质交联,氧化损伤是导致DNA损伤的原因之一。 Objective To study the DNA damage and its way in V79 cell caused by diesel exhaust particles extracts(DEPE). Methods Detected the DNA damage with single-cell gel electrophoresis(SCGE) ,and the V79 cell were pretreated with N-acetylcysteine (NAC) to explore the way of DNA damage. Results It showed that single strands break (SSB) and double strands break (DSB) were observed in V79 cell exposed to DEPE. Moreover, DEPE also could induce DNA-protein crosslinks (DPC)at dosage of 500 and 1 000 μg/mL;and pretreatment of NAC could reduce the toxicity of DEPE to the cells. Conclusion DEPE could cause V79 cell three types DNA damage,including SSB, DSB and DPC,and the DNA damages might be related to oxidative stress.
