食管癌组织3p、18q位点微卫星杂合性缺失的研究 被引量:3

摘要 目的探讨食管癌组织中3p14.3~3p21.1、18q22.3~18q23杂合性缺失(loss of heterozygosity, LOH)频率与人食管癌发生及其临床病理特征的关系.方法应用聚合酶链反应(polymerase chain reaction, PCR)和变性聚丙烯酰胺凝胶电泳技术(denaturalized polyacrylamide gel electrophoresis, PAGE),对28例食管癌中微卫星DNA序列不稳定性(microsatellite instability, MSI)表达情况进行研究.结果 28例食管癌标本中23例鳞状细胞癌9例LOH阳性,5例食管小细胞癌4例LOH阳性;LOH与肿瘤的病理分级、PTNM分期、有无区域淋巴结转移和浸润深度无相关性(P>0.05);3P位点上有较高频率的LOH发生.结论 LOH与食管癌的临床病理类型、临床分期、细胞分化程度、癌组织浸润深度和有无区域淋巴结转移等参数无相关性(P>0.05);食管癌在多个染色体位点均存在LOH现象,3p是食管癌相关的热点区域,3p位点基因的改变在食管癌发生过程中具有较重要意义.
