摘要
目的研究糖尿病大鼠肾组织中转化生长因子β1(transforming growth factor-β1,TGF-β1)和Smad4、Smad7的表达。方法用链脲佐菌素诱导的Wistar糖尿病大鼠为模型,随机分为正常对照组和糖尿病组,每组12只,于给药8周后处死,检测各组大鼠的平均动脉压、血糖、肾功能、24 h尿蛋白;采用逆转录多聚酶链反应(reversetranscription-polymerase chain reaction,RT-PCT)检测TGF-β1及Smad4、Smad7 mRNA表达水平;采用免疫组织化学法检测TGF-β1、Smad4和Smad7蛋白定位表达。结果与对照组比较,糖尿病大鼠平均动脉压、血糖、肾功能、24 h尿蛋白均增加,TGF-β1,Smad4和Smad7 mRNA表达上调,TGF-β1,Smad4和Smad7蛋白表达明显增加,且均有统计学意义(P均<0.01)。结论TGF-β/Smad通路在糖尿病肾病时是激活的。
Objective To investigate expression of transforming growth factor-β1 (TGF-β1 ), Smad4 and Smad7 in kidney tissiue of diabetic rats. Methods Healthy male Wistar rats were divided into two groups randomly, normal control group and strepotozocin induced diabetic rats group(12 rats in each group). The rats were sacrificed after drug administration for eight weeks. Mean arterial pressure, plasma glucose, kidney function, 24 h urinary protein were observed. The level of TGF-β1, Smad4 and Smad7mRNA were examined by reverse transcription-polymerase chain reaction(RT-PCR). The localization TGF-β1, Smad4 and Smad7 protein were examined by immunohistochemistry staining. Results Compared to normal control group, mean arterial pressure,plasma glucose, kidney function, 24 h urinary protein were significantly increased in diabetic rats group. The levels of TGF-β1 and Smad4,Smad7mRNA were up-regulated as well as the expression of TGF-β1 and Smad4, Smad7 protein. Conclusion TGF-β/Smad signaling is activated in diabetic nephrology.
