摘要
目的简便检测AmpC酶三维试验的方法。方法将提取AmpC酶冻溶的温度-70℃改为-20℃,将加酶的挖槽法改为打孔法。结果55株耐三代头孢菌素和头孢西丁的细菌,用两种冻溶方法均检出6株高产AmpC酶菌,其中用原提酶法,5株菌在挖槽和打孔中加入粗酶液10μl,另1株菌在挖槽和打孔中加入粗酶液30μl时出现阳性结果;用改进提酶法,5株菌在挖槽和打孔中加入粗酶液10μl,另1株菌在打孔中加入粗酶液30μl,而在挖槽中加入50μl时出现阳性结果。结论改进的提取AmpC酶的方法与原方法结果相同,改进的加酶打孔法比挖槽法优越,改进的三维试验方法简便、准确、不需要特殊设备,适合于各级实验室应用。
OBJECTIVE To simplify the three-dimensional test detection method of AmpC β-lactamase enzyme. METHODS The temperature of AmpC enzyme extraction and freezing-thawing was changed from -70℃ to -20℃. The method of adding enzyme is changed from slit to hole. RESULTS We detected 55 isolates which were resistant to the third generation cephalosporin and cefoxitin. It showed that 6 strains were both detected as hyperproducing AmpC enzyme by two methods. Among the 6 strains, 5 strains occurred positive result when 10μl crude enzyme liquid was added while the other one strain was positive when 30μl added by the old enzyme extraction method. Using the new enzyme extraction method, 5 strains was positive when 10μl added while the other one strain was positive when 30 μl added by hole and when 50μl added by slit. CONCLUSIONS The result of the improved AmpC enzyme three-dimensional test is consistent with the old one. The improved new threedimensional test is simple, correct, not demanding special equipments and fit for every grade laboratory.
