系统性尖端赛多孢子菌感染小鼠白细胞介素10表达的研究

摘要 目的:了解白细胞介素10(IL-10)在系统性尖端赛多孢子菌感染中的表达情况。方法:建立小鼠系统性尖端赛多孢子菌感染模型,包括致死量感染组(健康鼠5×106孢子尾静脉接种)、亚致死量感染组(健康鼠5×105孢子尾静脉接种)及免疫抑制组(地塞米松诱导免疫抑制鼠给予5×105孢子尾静脉接种),用逆转录—聚合酶链反应技术(RT-PCR)检测脾内白细胞介素10(IL-10)mRNA水平,用平皿系列稀释法检测肾内菌落形成单位(CFU),并记录平均生存时间(MST)。结果:致死量组中小鼠脾内IL-10表达明显上升(3 d,P<0.01;7 d,P<0.01),小鼠肾内大量菌生长,且7 d高于3 d(P<0.01),小鼠平均13.6 d均死亡;亚致死量感染组中,IL-10表达基本正常,肾内菌量少,且7 d低于3 d(P<0.01),小鼠生存时间均超过45d;与正常对照组相比,免疫抑制组小鼠脾内IL-10表达明显上升(3 d,P<0.01;7 d,P<0.01),小鼠肾内大量菌生长,且7 d高于3 d(P<0.01),小鼠平均14.2 d死亡,此外,免疫抑制组小鼠脾内IL-10及肾内菌量均高于相应时间亚致死量组(3 dP<0.01,7 dP<0.01:CFUs:3 dP<0.01,7 dP<0.01)。结论:IL-10在小鼠系统性尖端赛多孢子菌感染中表达增高,可能起着促进疾病发展的作用。 Objective : To investigate the role of lnterleukin - 10( IL - 10) in experimental murine of systemic scedosperium apiospermum infection. Methods: Three murine systemic scedosperium apiospermum infection models were created, including lethal doses group (5 × 10^6 spores were inoculated in intact mice by tail vein) , sublethal doses group( 5 × 10^5 spores were inoculated in intact mice by tail vein) and immunosuppressed group ( dexamethasone - induced immunosuppressed mice were inoculated with 5 × 10^5spores by tail vein). In these models, reverse transcription- polymerance chain reaction technique( RT- PCR) was applied to detect the expression of mRNA for IL - 10 in spleens, clone forming units (CFU) of fungi in the infected kidneys were determined with the plating dilution method, and mean survival time(MST) of the mice was also recorded. Results: In lethal doses group, the expression of IL - 10 mRNA increased markedly(3 d, P 〈0.01; 7 d, P 〈0. 01 ). There were a large amount of fungi in kindeys of lethal doses group and CFUs in kidneys on 7 days were higher significantly than on 3 days post - infected (P 〈 0. 01 ). The MST was 13.6 days for lethal infection group. In the sublethal doses group, the expression of mRNA for IL - 10 was within normal limits. There were a few fungi were found in kindeys of sublethal doses group and CFUs in kidneys on 7 days were lower significantly than on 3 days post- infected(P 〈 0.01 ). The MST was beyond 45 days for lethal infection group . Compared to normal controls, the expression of mRNA for IL - 10 increased markedly (3 d, P 〈0. 01 ; 7 d, P 〈0.01 ). There were a large amount of fungi in kindeys of immunosupressed group and CFU in kidneys on 7 days were higher significantly than on 3 days post - infected (P 〈 0.01 ). The MST was 14.2 days for immunosupressed group. Furthermore, the expression of mRNA for IL -10 and CFUs in immunosupressed group were significantly higher than those in sublethal doses group at the same time(3 d, P 〈0.01 ; 7 d, P 〈0.01 ; CFUs: 3 d, P 〈0.01, 7 d, P 〈0.01 ).
