摘要
目的:探讨雷公藤内脂醇对脓毒症大鼠血清炎症介质的干预作用。方法:90只SD大鼠随机分为3组,每组30只。B组盲肠结扎穿孔制造脓毒症模型;A组(假手术组)除不接扎盲肠,不刺穿盲肠外,余操作同B组;C组盲肠结扎穿孔后,立即腹腔注射0.05g/L的雷公藤内脂醇4ml/kg,并每隔8h追加1次。各组大鼠在术后3h、12h、24h分次处死,每次10只,取下腔静脉血,全自动生化检测仪检测血清丙氨酸氨基转移酶(ALT)和肌酐(Cr)水平,ELISA法测TNF-α、IL-1β水平,并取肝、肾组织行HE染色做病理检查。结果:与A组比较,B、C组大鼠血中ALT、Cr、TNF-α、IL-1β水平均升高,肝肾功能与形态出现损伤性变化;但C组血清炎症介质水平较B组降低,肝肾功能与形态损伤减轻。结论:雷公藤内脂醇可能通过减少机体炎症介质的释放,从而对脓毒症发挥治疗作用。
Aim : To investigate the effect of triptolide on the changes of inflammatory mediators in septic rats. Mothods: Ninety Wistar rats were randomly allocated into three groups(30 in each group) : sham operation group (group A) , group B and group C. Group B and group C were subjeted to make septic rat model,and then group C was given triptolide by intraperitoneal injection immediately. At 3 h, 12 h and 24 h after the establishment of model, 10 rats in each group were killed,respectively, and serum TNF-α, IL-1β,ALT and Cr were determined, and liver and kidney tissue was obtained to observe the pathological changes under microscope. Results: Compared with those of group A, serum TNF-α, IL-1β,ALT and Cr levels in group B and C were higher,and liver and kidney revealed morphological injury. All the parameters mentioned above in group C were significantly lower and the pathological changes were milder than those in group B. Conclusion : Triptolide is helpful to treat rat sepsis by down-regulating the levels of inflammatory mediators.
