摘要
目的:探讨温热和冬凌草液对体外培养的人膀胱癌BIU-87细胞体外凋亡的影响。方法:通过对体外培养的BIU-87细胞进行加温(37℃、41.5℃、43.5℃)和冬凌草液(0g/L、0.15g/L、0.3g/L、0.6g/L、1.2g/L、2.4g/L)处理,采用形态学和流式细胞术观察温热和冬凌草液诱导的细胞凋亡效应,采用MTT法检测冬凌草作用后细胞的生长抑制率。结果:冬凌草液对BIU-87细胞具有明显的抑制生长作用。形态学观察到实验组中大量具有凋亡特征的细胞。流式细胞仪定量分析显示,随着冬凌草液浓度的增加和温度的升高,BIU-87细胞凋亡率亦呈升高趋势(P<0.05)。结论:温热和冬凌草液具有协同诱导人膀胱癌BIU-87细胞凋亡的作用。
Aim ： To investigate the effect of hyperthermia and herba rabdosia rubescens extract on the apoptotic rates of bladder carcinoma BIU-87 cell line cultured in vitro. Methods ： The apoptotic rates of bladder carcinoma BIU-87 cell line cultured in vitro were determined with flow cytometry after being treated with hyperthermia （37℃ ,41.5℃ ,43.5℃ ） and herba rabdosia rubescens（0 g/L,0.15 g/L,0.3 g/L,0.6 g/L, 1.2 g/L,2.4 g/L） ,respectively, and the features of cell apoptosis were observed under microscope and transmission electron microscope;the ceil growth inhibition rate was determined using MTT. Results： The inhibitive effect of herba rabdosia rubescens on bladder carcinoma BIU-87 cell line was obvious. The cells with apoptotic characteristic was observed in the experimental groups, but not in the control group. Hyperthermia combined with herba rabdosia rubescens extract affected the apoptotic rates （P 〈 0.05）. Conclusion ： Hyperthermia and herba rabdosia rubescens extrac have coordination effects on inducing bladder carcinoma BIU-87 cell line apoptosis.
