低浓度三氯乙烯诱发L-02肝细胞蛋白质组异常表达

摘要 【目的】研究低剂量三氯乙烯对人L-02肝细胞蛋白质表达谱的影响,以助于阐明三氯乙烯引起细胞早期应答反应的分子机制。【方法】L-02肝细胞暴露于低剂量(3μmol/L)三氯乙烯24h后,提取细胞总蛋白,双向电泳分离蛋白质,软件分析凝胶图像,基质辅助激光解吸电离飞行时间串联质谱(MALDI-TOF-TOF-MS)对相关异常变化斑点进行鉴定。【结果】和对照组比较,低剂量三氯乙烯处理后L-02肝细胞蛋白质表达发生改变,初步鉴定出7个差异蛋白。低剂量三氯乙烯刺激后,上调的蛋白有核糖体样蛋白(similartoribosomalprotein)和SETprotein,下调的蛋白有异柠檬酸脱氢酶(isocitratedehydrogenase,NADP)和腺苷二磷酸-核糖基化因子鸟苷酸因子6(ADP-ribosylationfactorguaninenucleotidefactor6),微管-肌动蛋白交叉连接因子1(microtubule-actincrosslinkingfactor1)特异表达,肽基脯氨酰顺-反异构酶PPI(peptidylprolylcis/transisomerase)缺失。【结论】低剂量的三氯乙烯处理后,L-02肝细胞中的蛋白表达谱发生明显变化,这为三氯乙烯毒作用机制的进一步研究提供了线索。 [Objective] To investigate the effects of low concentration of trichloroethylene (TCE) on the expression of proteins in L-02 cells, and to study the possible molecular mechanisms of early cellular response induced by TCE. [ Methods ] After L-02 liver cells treated with 3 μmol/L TCE for 24 h, the proteomic expression of L-02 liver cells were analyzed by 2-D electrophoresis and the differentially expressed protein spots were identified by matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-TOF-MS). [Result] Seven changed proteins in L-02 liver cells treated with low TCE were identified by MALDI-TOF-TOF-MS. Among these 7 proteins, the expression of similar to ribosomal protein and SET protein was upregulated, while, isoeitrate dehydrogenase [NADP] and ADP-ribosylation factor guanine nucleotide factor 6 were downregulated; microtubuleactin crosslinking factor 1 was expressed specifically, peptidyl prolyl cis/trans isomerase disappeared. [ Conclusion ] The proteome expression appeared to be significantly changed in L-02 liver cells treated with low concentration of TCE, which would be a hint for further study of toxic mechanism of TCE.
