摘要
目的:揭示记忆是否在异相睡眠中得到加强和巩固,取消异相睡眠是否导致记忆维持能力下降及其下降的机制.神经元性一氧化氮合酶(neuronal n itric oxide synthase,nNOS)是否参与记忆中枢海马结构调节,剥夺异相睡眠(para-doxical sleep deprivation,PSD)是否改变它们在海马的表达.方法:①采用小平台法建立PSD大鼠动物模型.应用Morris水迷宫测试系统检测大鼠空间参考记忆获得及维持的变化;②海马的nNOS免疫组化研究:PSD 24,48和72 h后,立即取脑、固定、行冰冻切片(片厚25μm)、SABC法免疫组化染色、光镜下进行观察分析、摄像.结果:①PSD可造成大鼠空间参考记忆维持能力的损伤;②PSD组大鼠海马中nNOS免疫反应阳性细胞在CA1,CA2,CA3,CA4及锥体细胞层、颗粒层、齿状回稀疏,数目较相应的大平台组(tank control groups,TCgroup)、正常饲养组(control groups,CC group)明显减少(P<0.05).而TC组与CC组相比较无明显变化.结论:①PSD可以造成大鼠空间记忆的巩固障碍,异相睡眠与空间记忆维持有关;②PSD可以造成海马nNOS阳性细胞的表达减少,海马中的nNOS可能参与了大鼠空间记忆巩固的过程.
AIM: To study the changes of memory processes and consolidation during paradoxical sleep in rats and to observe the alteration of neuronal nitric oxide synthase immunoreactivity (nNOS-IR) neurons in hippocampus after paradoxical sleep deprivation. METHODS : Aradoxical sleep deprivation was performed using the “lower pot technique” The control groups (CC), tank control groups(TC) and paradoxical sleep deprivation groups(PSD) were tested in Morris Water Maze. At the end of the experiment, the animals were sacrificed under chloral hydrate anesthesia (0.4 g/kg) by intra-aortic perfusion of a fixative solution. Coronal sections were cut at 25 μm thickness on a freezing microtome and up to five series of adjacent sections were collected for nNOS immunohistochemical processing using streptavidin-biotin-peroxidase complex technique (SABC technique). RESULTS: The memory consolidation in PSD 24 h, PSD 48 h and PSD 72 h rats declined significantly compared with that in TC 24, 48, ?2 h as well as CC 24, 48, 72 h rats ( P〈0.05 ), indicating that PSD caused a significant impairment in reference memory. The nNOS-expressing cells in hippocampus of PSD 24, 48, 72 h rats were significantly fewer than those in TC 24, 48, 72 h as well as CC 24, 48, 72 h rats ( P 〈 0.0.5 ). CONCLUSION : ①Paradoxical sleep deprivation may cause a disturbance of memory processes and consolidation and paradoxical sleepis associated with memory. ②The nNOS-IR neurons in hippocampus formation decrease in PSD groups compared with those in CC and TC groups, nNOS in hippocampus may play a key role during paradoxical sleep and spatial memory.
