摘要
目的观察糖尿病(DM)大鼠股骨头缺血性坏死(ANFH)血浆内皮素(ET1)、前列腺素E2(PGE2)、血栓素B2(TXB2)、6酮前列腺素F1α(6KetoPGF1α)水平的变化,分析其在ANFH发病机制中的作用。方法SD大鼠腹腔内注射链脲佐菌素(STZ)60mg/kg建立速发型STZDM模型,测定DM大鼠及对照大鼠血浆ET1、PGE2、TXB2及6KetoPGF1α水平,观察股骨头病理学变化。结果DM组与对照组比较,ET1、PGE2、TXB2水平以及TXB2/6KetoPGF1α(T/K)比值均有显著升高,而6KetoPGF1α水平显著降低。结论血浆ET1、PGE2、TXB2以及T/K值的平衡失调可能参与并促使DM大鼠股骨头发生微血管病变,最终导致缺血坏死。
Objective To observe the changes in contents of plasmendothelin-1(ET-1), prostaglandin- E2 (PGE2), thromboxan B2 (TXB2) and 6-ketone-prostaglandin F1α (6-Keto-PGF1α) in the model rats of avascular necrosis of the femoral head(ANFH) induced by diabetes . Methods To establish immediate streptozotocin (STZ)-DM model rats using intraperitoneal injection one time with the dose of 60 mg/kg, then determined the levels of plasma ET-1, PGE2, TXB2 and 6-Keto-PGF1α of the model rats and observed the pathology of the femoral head. Results It was shown that the levels of plasma ET-1, PGE2, TXB2 in the experimental group all remarkably increased, while that of 6-Keto-PGF1α decreased strikingly comparing with the control. Conclusion The factors of plasma ET-1, PGE2, TXB2 and the dysequilibrium of T/K ratio involve in the development of microangiopathy in femoral head and elicit of ANFH.
