摘要
目的:以转染HBV全基因组并稳定分泌病毒抗原的HepG2.2.15肝癌细胞系为细胞模型,探索IFN-γ对HBV相关性肝癌的免疫调节效应。方法:用HBsAg和HBeAg ELISA检测试剂盒测定IFN-γ作用0,6,12,24h后,HBV病毒颗粒的分泌表达水平。通过MTT法检测不同剂量IFN-γ对新型凋亡配体TRAIL与TWEAK的生长抑制效应的调节作用,不同剂量IFN-γ对TRAIL与TWEAK的凋亡诱导效应的调节作用。结果:IFN-γ在不同的时间点对HBsAg和HBeAg的分泌均具有抑制作用,IFN-γ能够显著增强TRAIL和TWEAK对HepG2.2.15肝癌细胞系的生长抑制效应和凋亡率。讨论:IFN-γ是HBV感染后的一个重要的免疫效应分子,它可以通过抑制病毒复制和增强免疫监视分子的效应而在HBV相关性肝癌中发挥抗病毒和抗肿瘤效应。
Objcetive :The HBV transgenlc HepG2.2. 15 hepatocarclnoma cell llne was used as a cell model to explore the immune resulatlon effect of IFN-γ on the HBV related hepatocarcinoma. metkods ;The HBV particles secreted by HepG2.2. 15 cells were deteoted by HBsAg HBeAg detectlng ELISA kits. Modulation of IFN-γ to the the growth Inhibiton effeot Induced by apoptosis Induclng lisand TRAIL and TWEAK were assayed by MTT method. Modulatlon of IFN-γ to the apoptosis Inducing effeot of TRAIL and TWEAK were assayed Ioy flow cytometry. Resmlis : Seoretlon of HBsAg and HBeAg could be Inhibited by IFN-γ. The growth inhibition effect and apoiotosls induced by TRAIL and TWEAK could be greatly increased by IFN-γ. Coneluslon : IFN-γ Is a very important Immune effective molecule that will play a key role after HBV Infeotlon. It could exert its immune function by ;nhibiting the virus secretion and enhance the immune surveillance In HBV related hepatocarclnoma.
