摘要
以黑麦草成熟胚为外植体材料,对外植体的灭菌、愈伤组织的诱导、继代及植株再生及其过程中部分影响因素进行了研究.结果表明:用75%酒精对外植体处理60 s后,用75%NaClO原溶液(有效氯为7%~10%)灭菌25~30 min,灭菌效果最佳.基本培养基类型、植物生长调节剂对愈伤组织诱导均有影响,采用MS为基本培养基,添加2,4-D浓度为2.0~3.0 mg/L,6-BA浓度为0.1 mg/L时最高诱导率达86.67%.
The factors influencing plant regeneration of the callus from the mature embryo of Ryegrass which including explant sterilization, callus induction, subculture and plant regeneration was studied. The result showed that the explants, surface-sterilized by a solution of 75 % NaClO(7% - 10% active chlorine)for 25 - 30 min after dipped in 70% ethanlo for 1 min could get optimam sterilization effect. The feasible medium for induction of callus was MS. basal medium supplemented with 2.0 - 3.0 mg/L 2,4-D, 0.1 mg/L 6-BA, in which the rate of the callus induction reached to 86.67%.
