摘要
目的了解念珠菌属 DNA 合成周期各时相的构成。方法应用粘附式细胞仪、碘化丙啶染色法,对念珠菌细胞进行断层扫描,测其综合荧光值,比较研究白色念珠菌、热带念珠菌、假热带念珠菌和季也蒙念珠菌的 DNA 综合荧光强度和 DNA 合成周期各时相构成比。结果念珠菌细胞DNA 合成周期各时相构成比较,种间差异有显著性,同种不同株的白念珠菌间差异无显著性。热带念珠菌 DNA 综合荧光值最高,季也蒙念珠菌和白色念珠菌较低。结论念珠菌的 DNA 合成周期各时相构成在菌种鉴定方面可能具有一定的价值。
Objective Researches have been done in order to discover the component proportion of subacute period of DNA synthetic cycle of candida species.Methods DNA integrated fluorescence in- tensity of candida isolates including C.albicans,C.tropicalis,C.guilliermondii and C.pseudotropicalis growing in exponential-growth-phase was determined by adherent cell analysis and sorting cytometer. All samples were treated with RNAse and stained with propidium idodid(PI).Results The results showed that the component proportion of subacute period of DNA synthetic cycle was significant discrep- ancy between species,but interspecific difference in integrated fluorescence value was'nt found. Conclusions It's suggested that analysis of the proportion of G_1,S,G_2M phase in the DNA synthetic cycle may provide a new way of distinguishing fungus specimen.
