摘要
用反相高压液相色谱法检测43例大肠癌组织及癌旁粘膜中多胺(精脒、精胺)水平,结果发现:癌组织中精脒含量明显高于正常大肠粘膜(P<0.05),精脒/精胺比值明显高于正常大肠粘膜及癌旁粘膜(P<0.001);低分化癌高于高分化癌(P<0.05);Dukes B期及C期癌组织中精脒含量及精脒/精胺比值均明显高于A期(P<0.05),B期及C期癌组织中精脒/精胺比值亦明显高于同期癌旁粘膜(P<0.05)。表明精脒参与了大肠癌细胞的增殖和分化,精脒水平及精脒/精胺比值反映了大肠癌细胞的增殖活性,多胺代谢是大肠癌生长速度调控中的一个环节,为大肠癌病人研制与应用多胺抑制剂提供实验依据。
Using high-performance liquid chro- matographic techniques, the spermidine and spermine levels in tissue of large intestine car- cinoma (LIC) and its adjacent mucosas were measured in 43 patients. The results showed that the spermidine content in cancer tissue was obviously higher than that in normal mu- cosas (P<0.05), and the ratio of spermidine to spermine in cancer tissue was evidently higher than that in its adjacent mucosas and normal mucosa (P<0.001). And this ratio in carcinoma tissue of high histodifferentiation degrees was higher than that in carcinoma tis- sue of low histodifferentiation degrees (P< 0.05). Compared with cancer tissue of Duke's A stage, the spermidine level as well as the ra- tio of spermidine to spermine in cancer tissue of Duke's B and C stages was markedly higher (P<0.05 for both parameters). The ratio of spermidine to spermine in cancer tissue of Duke's B and C stages was significantly higher than that in their corresponding adjacent mu- cosas (P<0.05). These results showed that spermidine plays a role in the proliferation and differentiation of LIC cells, and the spermidine content and the ration of spermidine to sper- mine reflect the proliferation activity of LIC cells, and the metabolism of polyamine is one of the links related to the regulation of the growth speed of LIC cells. Our results provid- ed the experimental basis for the development and application of polyamine-inhibiting drugs.
