摘要
目的以我院临床分离的肺炎克雷伯菌KP1为研究对象,对其所产生的质粒AmpC酶编码基因的序列进行研究。方法采用聚合酶链反应(PCR),用DHA1型AmpC酶引物扩增KP1接合子中AmpC酶全编码基因,并克隆到pUC118载体中进行表达,对PCR产物进行测序,分析其基因型别。结果肺炎克雷伯菌KP1菌株的转移接合子所含有的质粒AmpC酶为DHA型,编码基因序列与GenBank中DHA1编码基因序列的同源性为100%。结论哈尔滨地区临床分离肺炎克雷伯菌有可转移性DHA1型AmpC酶的存在。
Objective To investigate the gene encoding the plasmid-mediated AmpC β-lactamase of a Klebsiella pnuemoniae strain isolated from a patient in the Second Hospital of Harbin Medical University. Methods AmpC β-lactamase gene in the transconjugate from Klebsiella pnuemoniae isolate was amplified by PCR, the purified PCR product was cloned into pUC118 vector and sequenced by dideoxy chain termination composition method. Results The encoding gene of the AmpCC-β-lactamase was identified as DHA-1 gene. Conclusion The DHA-1 AmpC β-lactamase exsists in Harbin.
