摘要
目的研究扇贝糖蛋白的蛋白及多糖的含量及组成。方法栉孔扇贝(Chlamys farreri)的扇贝边经水煮、分离纯化、醇沉及酶解等分离出扇贝酸性糖蛋白(FAGP)、扇贝中性糖蛋白(FNGP)、及扇贝多糖(FPS);通过纸层析(PC)、氨基酸分析、UV、IR、元素分析等技术确定其单糖及元素组成。结果PC分析FAGP单糖组成为:葡萄糖、甘露糖、鼠李糖、岩藻糖、半乳糖、木糖、葡萄糖醛酸。主要元素含量C42.62%、H5.73%、N6.81%。
OBJECTIVE To study the protein and glucose constituent of Chlamys farreri glycoprotein. METHODS FAGP,FNGP,FPS are prepared from Chlamy farreri scallop,by exaction with hot water,isolated and purified with column chromatography on DEAE-cellulose and sephadex G-100 successively. FPS is enzymed from FAGP. Sugar and Aa composition were determined by PC, Aa analysis, UV, IR, element analysis technology. RESULTS PC shows the sugar components of FAGP were:glucose, manose, rhamnose, fucose, galactose, xylose glucugonic. The protein and sacchride content of FAGP was 60%, 36 %respectively. The main elements contents were C42.62%,H5.73 %,N6.81%.
