摘要
目的:探讨天麻抑制平滑肌收缩的有效部位及作用机制。方法:离体平滑肌实验法。结果:天麻提取物G2表现了明显钙通道阻滞作用,其可浓度依赖性地使兔回肠平滑肌自律性收缩幅度下降,IC50值为0.102mg.ml-1;对CaCl2累积浓度-效应曲线产生非竞争性拮抗作用,且作用强度呈浓度依赖性增强;G2可浓度依赖性地抑制乙酰胆碱(Ach)所致的第二时相收缩。结论:天麻提取物G2是通过抑制细胞膜上的电压依赖性钙通道(PDC)而达到舒张离体平滑肌作用的。
Objective： To discuss the effect of isolated ileum smooth muscle and mechanism on tile extracting G2 from Gastrodia rhizome. Methods： The test of isolated smooth muscle was used to assay the mechanism of the relaxation. Resuits showed that G2 has inhibitory effect on distinctly calcium channel blockage action. The IC50 is 0. 102mg. ml^-1. G2 had non - competitive antagonistic action for CaC12 in an accumulated concentration - dependent manner. G2 also inhibited the second phase contraction induced by Ach. Conclusion： G2 had inhibitory effect on isolated ileum smooth muscles. The mechanism might be closely related to its inhibiting voltage dependent PDC on the cell membrane.
