自发性流产CBA/J×DBA/2小鼠母-胎界面DX5^+CD69^+细胞的浸润 被引量:2

Infiltration of DX5^+CD69^+ cells at the feto-maternal interface in spontaneous abortion CBA/J×DBA/2 mice

摘要 目的:研究DX5+CD69+细胞浸润与CBA/J×DBA/2小鼠自发性胚胎吸收的关系。方法:CBA/J×DBA/2小鼠(A组)和CBA/J×BALB/c小鼠(B组)分别用作自发性流产模型和生育力正常对照。DX5用作广泛性NK细胞标志物,CD69作为NK细胞和T细胞活化标志物。采用流式细胞术检测母-胎界面淋巴细胞中CD69+细胞百分率,DX5+CD69+细胞与DX5+细胞比率,并对细胞数量与胚胎吸收率进行线性相关回归分析。结果:A组孕13.5 d胚胎吸收率为34.1%(45/132),显著高于B组(6.6%,9/137;P<0.001)。同时A组DX5+CD69+细胞与DX5+细胞比率(28.1±11.9)%显著高于B组(19.5±5.7)%,(P<0.01)。此外,A组DX5+CD69+与DX5+细胞比率(x)和胚胎吸收率(y)之间存在强线性相关(r=0.807,P<0.001,y=-1.157+1.278x)。结论:母-胎界面过多的DX5+CD69+细胞浸润可能与CBA/J×DBA/2小鼠胚胎吸收有关。 Aim: To investigate whether DX5^+ CD69^+ cells are involved in recurrent spontaneous resorption of embryos in CBA/J female mouse mated with DBA/2 male (CBA/J × DBA/2). Methods: CBA/J × DBA/2 (group A) and CBA/J × BALB/c (group B) were used as abortion - prone model and fertile control, respectively. DX5 was used as a pan - NK cell marker, while CD69 was used as an activation marker on NK and T cells. The proportion of CD69^+ cells in lymphocyte group, and that of DX5^+ CD69^+/DX56+ at the feto- matemal interface were determined with flow cytometry. Linear correlation analysis was also performed between the cell number and resorption rate (RR). Results: RR was 34.1% (45 out of 132) in group A on day 13.5 of gestation. Such level was significantly higher than group B (6.6% ; 9 out of 137; P 〈 0.001). Similarly, the ratio of DX5^+ CD69^+/ DX5^+ cell was also significantly higher in group A (28.1 ± 11.9) % than in group B ( 19.5 ± 5.7) %, ( P 〈 0.01). In addition, a strong linear correlation was observed between DX5^+ CD69 ^+/DX5 ^+ cell ratio (x) and RR (y) (r =0.807, P 〈0.001, y = -1.157 + 1.278x)in group A. Conclusion: Infiltration of excessive DX5 ^+ CD69 ^+ cells at the feto - matemal interface may be associated with recurrent embryo resorption in CBA/J × DBA/2 mice.
