摘要
牛血样品通过低温冷冻、解冻及离心处理,除去细胞碎片后分成2份,1份为低水平,另1份加入Se^(+4)(高水平)。2份样品分别以2ml/瓶的量分装在棕色瓶中,冷冻干燥,辐射灭菌,最后于0℃以下保存。用荧光分光光度法测其均匀性与稳定性。结果本标准物质至少可以稳定1年。采用多个实验室多种方法确定的标准值为:低水平:(99.9±6.9)μg/L,高水平:(981±42)μg/L。
Samples of bovine blood were frosted, defrosted and centrifuged to destroy and separate blood cells. The blood was processed in two lots: the first lot was of low level, the second lot was fortified with Se^(+4) (high level) and homogenized. The two lots of blood samples were dispensed into brown glass bottles (2.0 ml per bottle), separately, and then lyophilized, radio-sterilized, and stored at less than 0℃. Homogeneity test, stability inspection and certification were conducted using a fluorescence spectrophotometric method. The physical and chemical stability of the CRM was assessed for one year. The certified values are based on analysis made by a group of independent laboratoriees using fluorescence spectrophotometric method, atomic absorption spectrophotometric method and atomic fluorescence spectrophotomdtric method. The certified values±uncertainties are as follows: Low level: 9.0±6.9μg/L, Elevated level: 981±42μg/L.
