摘要
以小鼠为模型,模拟阿霉素临床用药方式,研究具有过氧化物酶样活性的Ebselen的保护作用。通过测定心脏、肝脏、肾脏中脂质过氧化物(LipidPeroxides,LPO)含量、硒谷胱甘肽过氧化物酶(selenium-dependentglutathioneperoxidase,SeGSHPx)活性和血清中肌酸磷酸激酶(Greatinephosphokinase,CPk)活性及观察各脏器组织学变化,提示Ebselen能提高心肌抗氧化能力,即seGSHPx/LPO比值,降低LPO含量,防止和减轻阿霉素对心脏的脂质过氧化损伤。阿霉素对肝脏有轻微的组织学损伤,但完全可以受到Ebselen的保护。阿霉素对肾脏无明显的损伤作用。
driamycin-inducedsubacute cardio-toxicity occurs mostlikelyvia lipoperoxidative damage to tissues.Ebse-len is a selenorganic compound with Se-glutathione peroxidase-like activity.We tested this compound for its ef-fect on Adr-induced in vivo.The results demonstrared that Ebselen could decrease the content of LPO and ele-vate the ratio of SeGSHPx to LPO in heart tissue significantly.Correspondingly.the change of CPK in serum and the observation of the cross section of every tissue all showed that Ebselen could inhibit lipoperoxidative darnage from Adriamvcin efficiantly.
