摘要
在24 ℃和27 ℃恒温条件下孵化北草蜥(Takydromus septentrionalis)卵,以检测孵化过程中胚胎生长及物质的转化.从孵化第10天开始,每隔5天解剖卵,分离为胚胎、卵黄和卵壳.初生幼体称重后冰冻处死,随后解剖分离为躯干和剩余卵黄.所有样品65 ℃恒温干燥后称重.24 ℃和27 ℃北草蜥卵的平均孵化期分别为45.0天和32.1天.在24 ℃条件下,在卵孵化的最初30天内,胚胎仅动用了约35 %的卵黄物质,胚胎生长缓慢;孵化第30~45天,胚胎动用了约65 %的卵黄物质.在27 ℃条件下,孵化0~20天,胚胎动用了约28 %的卵黄物质;20~30天,动用了约72 %的卵黄物质.温度影响胚胎生长速率,但对胚胎生长模式的影响不显著,在前2/3孵化期,两种温度条件下的胚胎均生长缓慢, 而后1/3孵化期,胚胎生长则比较迅速.
A total of 213 eggs of northern grass lizards, Takydromus septenrionalis, were incubated at 24 and 27 ℃ using wet vermiculite as the incubation substrate, of which the moisture was kept at a constant of-220 kPa water potential. The growth trajectory of embryo and embryonic use of material were determined during incubation. From the tenth day of incubation, eggs were opened at 5-day intervals and separated into shell, embryo and yolk. The three egg components were oven dried to constant mass at 65℃, weighed and preserved frozen for later determination of composition. Upon emergence, the size and mass of each hatchling were measured and them killed by freezing to (15℃. Then each hatchling was separated into carcass and residual yolk fat bodies. The two components were oven dried to constant mass at 65℃, weighed and preserved frozen for later determination of composition. The incubation length averaged 45.0 and 32.1 days at 24 and 27 ℃, respectively. Embryos grew slowly during the first two third period of incubation duration independent of incubation temperatures, as indicatedby the small increase in embryo mass and the low rate of embryonic use of material.
