摘要
研究建立了一套适于我国北方马铃薯主栽品种东农303外源基因导入的体系,并成功地将烟草病原蛋白基因导入该品种.结果显示,块茎外植体比其它外植体更适用于基因导入。农杆菌浓度对T—DNA转移有明显影响,5×108CFU/mL是较适宜的浓度.组织化学和分子印迹杂交证明,外源基因已经整合入马铃薯染色体组中。试验选出8个转基因个体,将通过无性系繁殖,进一步鉴定遗传稳定性。
In search of establishing a system for genetic tansformation of potato, a pathogenesis-related protein gene from tobacco was successfully transferred into potato cultivar Dong Nong 303 via binary vector. According to southern blot analysis and histochemical assay for-glucuronidase, it was concluded that foreign genes was integrated in potato genome and could be expressed in potato tissues. Of three kinds of explants, a highly efficient introduction was obtained from tuber disc.The concentration of Agrbobacterium tumefaciens had a significent interaction on gene transfer. The most suitable concentration was 5 ×108CFU / mL. From present experiment 8 individuals transferred with foreign gene were selectedand expected to estimate genetic stability using somaclones.
