摘要
目的对HSV-1作抗原的玻片酶免疫法(EIA)用于B病毒抗体检测的可靠性进行研究。方法用HSV-1为抗原的玻片酶免疫法(EIA)和酶联免疫吸附法(ELISA)与B病毒为抗原的斑点酶免疫法(DIA)、免疫荧光法(IFA)及酶联免疫吸附法(ELISA)对猕猴血清进行B病毒抗体检测的比较分析。结果HSV1为抗原的EIA与B病毒为抗原的DIA、IFA和ELISA检测结果的符合率分别为100%、95.6%和97.4%;HSV1为抗原的EIA检测确定的阴性血清样本,经B病毒抗原的IFA检测均为阴性;在HSV1为抗原的EIA确定的779份阴性血清中,经B病毒抗原的ELISA检测,有2.6%(20份)为阳性;HSV-1抗原的EIA和HSV-1为抗原的ELISA检测均为阴性的20份血清样本,经B病毒抗原的ELISA检测,18份仍为检出阴性,2份为阳性。结论HSV-1为抗原的EIA的检测结果与B病毒抗原的DIA、IFA和ELISA的检测结果一致性较好。同时,用HSV-1抗原进行B病毒抗体检测,存在极少数阳性动物漏检的可能。
Objective To evaluate the reliability of enzyme immunoassay (EIA) of HSV-1 as antigen for detection of B virus antibodies. Methods B virus antibodies were detected with enzyme immunoassay (EIA, HSV-lantigen), enzyme-linked immunosorbent assay (ELISA, HSV- 1 antigen), immunobinding assay ( DIA , B virus antigen), immunofluorecenee assay (IFA, B virus antigen) and enzyme-linked immunosorbent assay (ELISA, B virus antigen) in macaque monkey sera. The testing results were compared and analysed. Results Consistency of testing results of EIA (HSV-1 antigen)with DIA ( B virus antigen), IFA (B virus antigen) and ELISA (B virus antigen)were 100%, 95.6% and 97.4%, respectively. The negative sera which were confirmed by EIA (HSV-lantigen) were still negative by IFA( B virus antigen). Among 779 negative sera confirmed by EIA (HSV-1 antigen), 2.6 % (20) was positive by ELISA (B virus antigen). Conclusion There is much consistency of testing results of EIA( HSV-1 antigen)with DIA (B virus antigen), IFA( B virus antigen)and ELISA (B virus antigen). Meanwhile, among B virus free animals which were confirmed using HSV-1antigen, very few animals would be positive using B virus antigen.
