摘要
目的研究子痫前期患者外周血单个核细胞分泌Th1/Th2细胞因子功能变化。方法子痫前期患者29例,正常妊娠妇女15例,分离外周血单个核细胞,体外培养72小时,ELISA方法检测培养上清液中Th1型细胞因子IL-2、IFN-γ及Th2型细胞因子IL-4、IL-10水平。结果子痫前期组PBMC培养上清液中,Th1型细胞因子IL-2、IFN-γ水平(259.4±90.5)pg/mL,(452.0±122.6)pg/mL与正常妊娠组(140.3±73.2)pg/mL,(307.5±106.4)pg/mL相比,显著升高,分别为P<0.01,P<0.05;Th2型细胞因子IL-4、IL-10水平(28.8±7.7)pg/mL,(178.9±98.6)pg/mL与正常妊娠组(41.9±11.4)pg/mL,(316.1±284.6)pg/mL相比,显著下降,分别为P<0.01,P<0.05。结论子痫前期患者外周血单个核细胞分泌Th1型细胞因子功能增强,分泌Th2型细胞因子功能减弱。
[Objective] To study the Th1/Th2 immunity in preeclampsia. [Methods] 29 patients with preeclampsia and 15 healthy pregnant women were selected by random, peripheral blood mononuclear ceils (PBMCs) were separated and cultured for 72 hours. Th1-type cytokines (IL-2 and IFN-γ) and Th2-type cytokines (IL-4 and IL-10) produced by PBMCs were detected by ELISA. [Results] In preeclamptic patients, concentrations of IL-2 (259.4±90.5) pg/mL in PBMC cultural supematants were significantly higher than in normal pregnant women (140.3± 73.2) pg/mL (P 〈0.01). and IFN-γ also increased significantly (452.0±122.6) pg/mL versus (307.5±106.4) pg/mL, P 〈0.05). Concentrations of IL-4 and IL-10were significantly decreased in preeclamptic patients than in normal pregnant women. (28.8±7.7) pg/mL versus (41.9±11.4) pg/mL, P 〈0.01, and (178.9±98.6) pg/mL versus (316.1±284.6) pg/ mL, P 〈0.05, respectively. [Conclusion] There is Th1/Th2 imbalance with increased Thl-type immunity and decreased Th2 -type immunity in preeclampsia.
