摘要
在裂殖酵母基本培养基的基础上,通过正交设计.优化了培养基6个组分;优化后的培养基,摇瓶发酵,目的蛋白产量在26-51 mg/L之间,发酵罐的批次发酵,产量在51-102 mg/L之间。
To get optimized ferment media of recombinant thanatin expressed in Schizosaccharomyces pombe, six components of EMM media, fission yeast minimal media, were studied by orthogonal experiment design. In optimized media, theoretical output of expressed thanatin can be maintained at 20-50 mg/L in flask, with 50-100 mg/L in fermenter. The key to get highly expressed recompinant thanatin in fission yeast is the control of initial media pH value and growth velocity.
