摘要
目的:探讨不同浓度的阿托伐他汀(atorvastatin)对氧化低密度脂蛋白(ox-LDL)诱导的人脐静脉内皮(HUVECs)细胞间黏附分子-1(ICAM-1)mRNA表达的影响.方法:体外培养人脐静脉内皮细胞,待细胞生长到融合状态时加入ox-LDL(100μg/ml),作用24小时后分别加入不同浓度的阿托伐他汀(1~30μmol/L),采用逆转录聚合酶链反应(RT-PCR)技术测定ICAM-1 mRNA的表达.结果:Ox-LDL可诱导ICAM-1 mRNA的表达,不同浓度的阿托伐他汀可抑制ox-LDL诱导的ICAM-1 mRNA的表达,并呈浓度依赖性(P<0.05).结论:阿托伐他汀可抑制ox-LDL诱导的HUVECsICAM-1 mRNA的表达,并呈浓度依赖性.
Objective: This study was to investigate the effects of atorvastatin treatment on intereelluar adhesion molecule - 1 (ICAM - 1 ), in cultured human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were cultured in endothelial basal medium.Cells were grown to confluence and used for oxidized low - density lipoproteins (ox - LDL) .After 24 hours,HUVECs were cocultured with the ox - LDL and atorvastatin( 1 - 30μmol/L) for another 24 hours. Expression of ICAM - 1 mRNA were determinded by reverse transcription - polymeras chain reaction (RT - PCR) . Results: Expression of ICAM - 1 mRNA were detected in ox - LDL - induced HUVECs. Atorvastatin dose - dependently ( 1 - 30μmol/L) decreased the ctpression of ICAM - 1 mRNA in ox - LDL- induced HUVECs, P 〈 0.05. Conclusion: Atorvastatin decreases dose- dependently the expression of ICAM- 1 mRNA in ox - LDL- induced HUVECs. Atorvastatin has anti - inflammatory effects which are probabiy related to reducing effect of ICAM - 1.
