The influence of bFGF on tbe biological characteristics of osteoblasts and periodontal ligament fibroblasts

摘要 目的:明确bFGF对体外成骨细胞和牙周膜成纤维细胞迁移、矿化相关蛋白合成的影响,以探讨bFGF促进牙周组织再生的内在机制以及在牙种植体组织界面局部应用bFGF诱导类牙周膜形成的可行性。方法:同一只SD大鼠来源的两种细胞经传代培养至第四代,建立细胞迁移模型,分别在普通培养基和含bFGF的培养基中培养,观察测量细胞迁移速度,并分别测试两种细胞在普通培养基和含bFGF的培养基中的碱性磷酸酶(ALP)活性及I型胶原的含量。结果:普通培养基培养时,成骨细胞迁移速度快于成纤维细胞;但牙周膜成纤维细胞加bFGF培养迁移速度明显高于其他各组;bFGF抑制成骨细胞的I型胶原的合成和ALP的活性。结论:bFGF能明显促进牙周膜成纤维细胞的移行;bFGF能抑制成骨细胞的碱性磷酸酶活性。 Objective: In order to determine the influence of bFGF on the biological characteristics of osteoblasts and periodontal ligament cells, thc authors investigated the influence ofbFGF on the migration and expression of protein related to mineralization of osteoblasts and periodontal ligament fibroblasts. Methods: The fourth passages of osteoblasts and periodontal ligament fibroblast were obtained and the wounding models in vitro of them were founded. The cells were cultured in mediums with bFGF or not. Their migration was observed and measuered. The ALP and Col-Ⅰ of these two kinds of cells cultured were evaluated. Results: The result showed that osteoblasts migrated faster than fibroblasts in common culture medium without bFGF and the periodontal ligament fibroblasts in culture medium with bFGF had higher migration rate than other groups. The Col-Ⅰ expression and ALP activity of osteoblasts were inhibited by bFGF. Conclusion: It is concluded that bFGF could accelerate periodontal fibroblasts' migration and bFGF could inhibit the ALPase activity of osteoblast.
