Glucan在Con A所致免疫性肝损伤中的作用

摘要 目的观察Glucan对刀豆蛋白A(ConA)所致小鼠免疫性肝损伤的干预效果及其作用机制。方法ConA尾静脉注射制备小鼠免疫性肝损伤模型,在ConA注射前1h腹腔注射Glucan,观察正常对照组、ConA损伤组、Glucan/ConA组肝损伤的情况,检测血清谷丙转氨酶(ALT)、α肿瘤坏死因子(TNF-α)及组织中一氧化氮(NO)含量的变化,并用DNAladder检测肝细胞凋亡的变化。结果ConA尾静脉注射后,出现明显的肝损伤,血清ALT、TNF-α及组织中NO水平明显升高,DNAladder检测可见明显的梯状条带;预先应用Glucan后,可减轻这些改变。结论Glu-can对ConA所致小鼠免疫性肝损伤有保护作用。 Purpose To observe the effect and the mechanism of Glucan on concanavalin A (Con A)-induced liver injury. Methods In Con A group, Con A was administered iv via tail vein. In Glucan/Con A group, mice were pretreated with Glucan prior to the injury of Con A. The ALT, TNF-α levels in serum and the nitric oxide (NO) levels in liver tissue were determined in control and the treated groups. Apoptosis was determined by DNA ladder detection. Results ALT, TNF-α levels in serum and the nitric oxide levels in liver tissue increased significantly after Con A injection. DNA ladder can be found. Pretreatment with Glucan could prevent these changes and lessen the Con A-induced liver injury. Conclusion Glucan has protective effect on Con A-induced liver injury.
