摘要
目的探讨β-联蛋白与结肠癌发生、发展以及转移的关系。方法利用组织芯片技术,采用免疫组化(S P)法检测62例结肠癌、18例结肠腺瘤和10例正常结肠组织中β-联蛋白(β-catenin)基因蛋白产物的表达水平。结果结肠癌和结肠腺瘤组织中β-catenin的异常表达率分别为80.6%和83.3%,均显著高于正常组织的表达率0.0%, 差异有显著性(P<0.01)。腺癌组与腺瘤组异常表达率差异无显著性(P>0.05)。低分化组异常表达率(87.2%) 明显高于高分化组(60.0%)(P<0.05)。淋巴结转移组异常表达率(92.5%)显著高于无淋巴结转移组(59.0%)(P <0.01)。β-catenin异常表达与肿瘤浸润深度和肿瘤大小无关(P>0.05)。结论β-catenin的异常表达与结肠癌的分化和转移有关,是肿瘤发生、发展及转移的一个重要因素,可作为结肠癌是否具有恶性潜能的一个重要参考指标。
Objective To study the expression and clinical significance of β-catenin in colon carcinoma. Methods Specimens from colon carcinoma (62 cases), colonadenoma ( 18 cases) and normal colon tissues ( 10 cases) for which tissue microarrays were constructed and used for immunohistochemical detection of β-catenin protein expression. Results Immunohistochemical analysis of tissue microarrays demonstrated β-catenin protein expression in 80.6 % of the colon carcinoma and in 83.3 % of colon adenoma. The expression of β-catenin was higher than that of normal tissues( P 〈 0.01 ), while between colon carcinoma and colon adenoma, being of no significance ( P 〉 0.05). β-catenin expression was significantly correlated with tumor differentiation status. Tumors with regional nodal metastasis showed higher abnormal staining rate (92.5 % ) than those without nodal involvement(59.0% )( P 〈 0.01).The expression of β-catenin was not relevant to depth or size of tumors. Condusion Abnormal expression of β-catenin is associated with invasion and metastasis of colon carcinoma, and is an important factor in development of highly invasive tumor. β-catenin could be used as an important marker of colon carcinoma.
