摘要
目的探讨MMP2、MMP9在大鼠局灶性脑缺血再灌注损伤时表达状况及其在损伤机制中的作用。方法建立大鼠大脑中动脉缺血2小时再灌注22小时模型,分别采用HE及SP免疫组化法观察大鼠全脑组织病理变化及MMP2、MMP9表达状况。结果假手术组大鼠脑组织形态结构正常,MMP2、MMP9见少量散在阳性表达或不表达;模型组大鼠大脑缺血区组织明显水肿、退变、坏死伴少量炎细胞浸润,对侧大脑相应区及双侧小脑轻度水肿,MMP2、MMP9表达较假手术组对应区显著升高,P<0.05或P<0.01,其中缺血侧大脑以坏死区边缘脑组织表达为甚,且较对侧大脑及双侧小脑明显升高,P<0.05。结论MMP2、MMP9参与了局灶性脑缺血再灌注损伤时包括病灶在内的全脑损害,且可能与临床上神经机能联系不能发生机制有关。
Objective To investigate the effects of the expression of MMP-2 and MMP-9 incerebral tissues of Rat after focal cerebral ischemia and reperfusion. Methods 2 hours right intraluminal middle cerebral artery occlusion with reperfusion was induced in 15 rats,and the specimen of brain were performed at the point after 22 hours reperfusion; histologic changes were observed by HE stained;expressions of MMP-2, MMP-9 were detected by S-P immunohistochemistry method. Results Both expressions of MMP-9 and MMP-2 were seen sparsely in the control specimens, and were markedly increased after transient focal cerebral ischemia and reperfusion( P 〈 0.05). Expression of MMP-2 and MMP-9 as well as vasogenic edema can be observed not only in the lesion itself( P 〈 0.01)but also in perilesional (penumbra),widespread ipsi- and contralateral remote areas (P 〈 0.05). Conclusion Expression of MMP-2 and MMP-9 is a significant response to transient focal cerebral ischemia/reperfusion and contributes to the early formation of vasogenic edema that can be observed all parts of brains. This may be involving the mechanism of diaschisis.
