摘要
目的建立大鼠骨髓间质干细胞(mesenchymalstemcells,MSCs)分离及培养的方法,探讨体外培养MSCs的生物学特性。方法通过全骨髓贴壁法分离培养大鼠MSCs,镜下连续观察细胞的形态变化。应用流式细胞仪测定细胞周期,并研究其增殖及生长特征。结果原代及传代培养显示,10代以前的MSCs具有活跃的增殖能力,细胞周期分析显示有16.8%的MSCs处于S+G2/M期。MSCs细胞形态可呈梭形、圆形或椭圆形,经传代融合时呈漩涡状、菊花状排列。结论体外培养10代以前的MSCs生长稳定,增殖较快,为进一步开展中枢神经系统疾病的移植治疗提供了细胞来源。
Objective To establish a method of isolation and cultivation of rat bone marrow mesenchymal stem cells(MSCs) in vitro and investigate the biological characteristics of MSCs. Methods Rat MSCs were isolated and cultured in DMEM. The morphology of MSCs was observed with microscope constantly. Cell cycle was analyzed by flow cytometry. The growth characteristic of MSCs was measured in passage cultures. Results MSCs showed active proliferative ability in passage cultere before the 10th. Cell cycle analysis showed that 16.8 % of MSCs was in S phase and G2/M phase. Conclusion Before the 10th passages,MSCs showed stable and rapid growth in vitro,MSCs could be used as the “seed cells” for treatment of central nervous system disease.
