摘要
目的探讨乙型肝炎表面抗原(HBsAg)假阴性的产生及预防方法。方法应用全自动加样仪(ML-AT plus-Sampler),使加样针在不同的清洗次数下,同时加样12份含不同浓度HBsAb标本和12份含不同浓度HBsAg不同OD值标本,分别应用ELISA二步法检测HBsAg,观察其吸光度的变化。结果 12份含HBsAg标本在清洗次数逐渐减少的情况下,OD值都有不同程度的下降。HBsAb强阳性标本对OD值影响很大,在只清洗3次的情况下,HBsAg弱阳性标本开始出现假阴性;在不洗涤也不擦拭的情况下,4份HBsAg弱阳性出现假阴性。结论高浓度的乙型肝炎表面抗体会引起HBsAg检测结果假阴性;使用一次性加样枪头,可避免假阴性。
Objective To study the reason and prevention of HBsAg false negative.Method 12 HBsAb positive specimens with different spectrophotometry(OD) and 12 HBsAg with different OD were aspired by ML-AT plus-Sampler with different times of tip cleaning and HBsAg was detected by two-step ELISA method.The changes of positive rate were observed in different times of tips cleaning.Results In the case for the times of clean tips gradually reduced,the OD values decreased in varied degrees.And the strong HBsAb positive samples had a great influence on the OD value.In the case of cleaning only for three times,HBsAg weak positive specimens appeared false negative.Four HBsAg weak positive specimens became false negative without wiping and washing.Conclusion High level of HBsAb will cause a false negative in HBsAg detection.Using one-time assay tips can solve the problem.
