摘要
浅麻醉Wistar大鼠49只,用辐射热照尾部作为伤害性刺激,以痛兴奋神经元(painexcitationneuronsPEN)诱发放电频率减少、痛抑制神经元(paininhibitionneuronsPIN)诱发放电频率增加和甩尾反射潜伏期(tailflicklatencyTFL)延长为镇痛效应,观察脑室注射不同剂量吗啡对尾核中PEN、PIN及TFL的同时影响。表明,脑室注射50μg/10μl吗啡,尾核中PEN、PIN放电呈镇痛效应(P<0.05),TFL影响不显著(P>0.05)。注射70、100μg/10μl吗啡,尾核中PEN、PIN放电和TFL均出现显著镇痛作用,且呈量效关系;吗啡对TFL的作用高峰晚于对尾核中PEN、PIN放电的作用,对TFL影响时间长。说明,吗啡对大鼠尾核中PEN、PIN两者的影响先于其对甩尾反射抑制效应的出现。
The effects of different doses of morphine administered to the lateral ventricle on firing of pain excitation neurons(PEN)and pain inhibition neurons(PIN) and tail flick latency(TFL) were observed simultaneously on forty nine lightly anesthetized Wistar rats with the radiant heat as noxious stimulation.The decrease of firing rate of PEN and the increase of firing rate of PIN and the Prolongation of TFL were regarded as antinociceptive effect.The results showed that the TFL did not change significantly while antinociceptive effect already appeared in PEN and PIN,when lower dosage(50μg/10μl) of morphine was used. However, when the dosage of morphine increased to 70, 100μg/10μl, the TFL increased significantly as well as the change in PEN and PIN, although the peak of the change in TFL appeared later and the effect on TFL lasted longer.The results suggest that the influence of morphine could be detected in both PEN and PIN in the caudate nucleus before its behavioral effect emerges.
